Two drops, warmed between the fingers, vanish into the skin within seconds. By morning, the mirror shows softer lines, a plumper cheek, a more rested face.
Estée Lauder prices that result at £65 for 30 millilitres. The promise is printed plainly: the serum “repairs the look of past damage and helps prevent future damage – in just one night”. Forty years of what the brand calls Night Science Research stand behind it. But can a serum applied to the surface reach the genes it claims to reset?
Between it and those genes stands the stratum corneum – the skin’s outer wall, built to keep almost everything out.
Javon Ford, an independent cosmetic chemist, describes the wall’s fixed rule. “If a product has a molecular weight of more than 500 daltons,” he says, “it won’t be able to penetrate the epidermis.” A dalton measures molecular size; above roughly 500, the door stays shut.
Estée Lauder calls Tripeptide-32 an “exclusive, patented 1-in-60 million” molecule – a cellular messenger that resets the skin’s internal clock. The brand says it reactivates the genes that govern overnight repair.
The science it invokes holds up. Skin does run on a circadian rhythm – a 24-hour cycle of daytime defence and night-time repair. Dr Derrick Phillips, a consultant dermatologist, sets out the division: “During the day, skin focuses on defence… At night, it switches into repair mode.”

Estée Lauder holds patents showing Tripeptide-32 switching those genes on. But the results come from cell cultures – skin cells in a dish, not skin on a face. No trial in the material reviewed for this investigation has reproduced the effect in living human skin.
Ford explains why the marketing language stays careful: “If the ingredients penetrate the skin, then that’s a drug claim. So even if these peptides were able to do that, they can’t make that claim unless they’re available by prescription.”
The serum’s base is thick with humectants – ingredients that pull water into the skin and hold it there. Glycerin, hyaluronic acid and several glycols flood the surface, swelling the cells and smoothing fine lines by morning. The effect is real and immediate. It is also temporary.
The chronobiology is real. Estée Lauder has not shown that its serum reaches the genes it claims to switch on.
543 women make up Estée Lauder’s largest test group. None of them were measured; they were asked, after a week and after a month, how their skin felt.
The objective claims rest on small panels: a measured 14 per cent reduction in fine lines, from 26 women, tracked twelve hours after one application. Estée Lauder lists Tripeptide-32 on the label without saying how much the bottle contains. No one outside the company can confirm whether today’s serum matches the concentration shown in its own patents.
A clinical trial numbered NCT03505684 is cited across beauty databases as proof that Tripeptide-32 cuts fine lines by 31 per cent. The registry, however, tells a different story: fish collagen, swallowed in capsules, by women in South Korea who never touched a serum.
